[求救] 想問一下IHC的二抗與blocking serum
我在Current Protocols in Neuroscience上
研讀IHC時
看到有句話這麼寫:
Nonspecific sites are preblocked with normal animal serum (NAS)
from the species in whick the secondary antibody was made.
意思是說
二抗跟normal animal serum要用同一個物種來源
我不懂為什麼得同一種
有問過其他前輩
有些人說沒聽說過 也沒有特別這樣用
也查不到為什麼
以上是小妹的第一個問題
另外
有到其他系所看別人作free floating(一種IHC的方法)
他們當時用脫脂奶粉作blocking
一抗是: rabbit anti-mice
二抗是: goat anti-rabbit
我當時不知道IHC也可以用脫脂奶粉blocking
於是請教他
甚麼樣的情況下可以不用NAS 而用脫脂奶粉
但他回答不上來
我自己也是查不到答案
